چکیده

Several reports exist which link low levels of serum alpha-1 antitrypsin with pulmonary emphysema. 3 ’9 ’13 Deficiency of this protein may be detected by serum electrophoresis or specifically quantitated by radial immunodiffusion. Recent investigations have led to the development of a number of automated and manual procedures for the quantitation of immunochemical reactions in aqueous media through the use of light-scattering techniques. 1 ’2 ’4 ’5 ’ 6 ’8 ’ 1 0 ’ 1 1 ’ 14 The present report describes a sensitive microtechnique for the nephelometric mea surement of the reaction between human serum alpha1 antitrypsin and its specific antibody.

Simple method for alpha1-antitrypsin deficiency screening by use of dried blood spot specimens.

The use of dried blood spot (DBS) specimens in quantitative alpha1-antitrypsin (alpha1-AT) detection or genetic analysis is limited because protein levels in the samples are low and they contain components that can interfere with polymerase chain reaction amplification. A methodological adaptation was developed to overcome these drawbacks which is discussed here. The study population consisted ...

Determination of Alpha 1-Antitrypsin Phenotypes and Genotypes in Iranian Patients

Alpha 1-antitrypsin (AAT) or alpha 1-protease inhibitor (PI) is the principal inhibitor of proteolytic enzyme in serum. Its phenotypic variability has been reported to be associated with liver, lung diseases and rheumatoid arthritis in humans. There is much documentation about high risk phenotypes of PI in some regions of the world, however, there are no reliable reports on these phenotypes and...

Serum Alpha-1-antitrypsin Evaluation by Three Electrophoretic, Enzymatic and Immunodiffussion Methods

Abstract Background & Objective: Alpha-1-antitrypsin (AAT) is the major component of the human plasma alpha-1 globulin proteins and acts as a major inhibitor of proteolytic enzymes, particularly elastase. AAT deficiency is accompanied by lung, liver and other disorders, therefore, AAT is clinically important and its precise evaluation is diagnostically critical. In present study serum AAT was e...

Luminometric assays of seven acute-phase proteins in minimal volumes of serum, plasma, sputum, and bronchioalveolar lavage.

We describe immunoluminometric assays for seven acute-phase proteins, which can be determined in minimal volumes of plasma, serum, sputum, and bronchioalveolar lavage. The theoretical volume of serum or plasma required to measure all seven analytes in duplicate is 130 nL, although in practice the smallest volume of sample was enough to fill a hematocrit tube (about 25 microL of blood), collecte...

Optimizing reference values for the measurement of alpha 1-antitrypsin in serum: comparison of three methods.

We studied three methods (rate nephelometry, radial immunodiffusion, and trypsin-inhibitory capacity) for their ability to detect those individuals with a deficiency of alpha1-antitrypsin. The phenotype represented in 170 serum samples was determined by isoelectric focusing as the reference method. All three methods correctly identified Pi Z, Pi S, and Pi SZ phenotypes but varied in their abili...
